Spectrum of Coverage:
- Haemophilus Influenza
- Neisseria meningitidis
- Streptococcus pneumoniae
- Salmonella enterica serotype Typhi
- Rickettsia
- Lymphogranuloma-psittacosis group
- Escherichia coli
- Staphylococcus aureus
Mechanism of Action:
Works by inhibiting protein synthesis by binding to the 50S ribosomal subunit & directly preventing the formation of bacterial protein. On a molecular level, chloramphenicol inhibits the attachment of transfer RNA to the A site on the 50S ribosome.
Indications:
- Rickettsial Diseases
- Meningitis
- Typhoid Fever
- Cholera
Dosage & Administration:
- Moderate Infections: 50mg/kg/day, given in 4 divided doses at 6 hours interval.
- Severe Infections (Meningitis & Bacteraemia): 100mg/kg/day, given in 4 divided doses at 6 hours interval with an attempt for dose reductions back to 50mg/kg/day made as soon as possible. Also, dose reductions to 25mg/kg/day may be necessary for neonates, and patients who have impaired liver or renal function.
